Abstract
Eggs
dissected from the ovaries of Hammerschmidtiella diesingi were incubated in
Locke's solution containing 0.1% (w/v) DL-ascorbic acid. The presence of
ascorbic acid enhanced embryonic development, significantly so between 8 and 42
h of incubation. Thereafter the cultures were similar to those which lacked
ascorbic acid. At 36 h, 60% of the eggs incubated with ascorbic acid were at
the active embryonic stage, whereas in control cultures at this time only 10%
were at this stage and 60% were at the pre-active embryonic stage.

In
chemotherapeutic treatment of helminthiasis, sensitivity of parasite to anthelmintic
drugs depends to a large extent on parasite's metabolic pathways, which may be
primarily either aerobic or anaerobic. But it has been difficult to understand
the ability of some of the worms to derive energy aerobically and
anaerobically. Anthelmintic drugs affect particularly carbohydrate metabolism
which holds the key role as the source of energy in helminth parasites. Studies
on carbohydrate metabolism of helminths have shown 2 main metabolic pathways.
One pathway shows predominance of glycolytic, as the entire source of energy;
in the other, the glycolytic pathway is shuttled at phosphoenol pyruvate stage
to oxaloacetate and malate through CO2 fixation. Some biochemical features of
carbohydrate metabolism 11 parasites were studied. A simple invertebrate
host-parasite model of Periplaneta americana and its nematode parasite, H.
diesingi was developed for the preliminary studies. The O2 uptake study on H.
diesingi was carried out by using Warburg manometric techniques. Intact worms
and homogenates were incubated for 3-8 h in glucose, pyruvate and acetate
substrates. The O2 uptake was equivalent with these substrates. In sugars, more
in galactose than glucose and mannose was found. The significance of these
results were discussed.
